{"id":136,"date":"2019-10-02T14:47:13","date_gmt":"2019-10-02T14:47:13","guid":{"rendered":"http:\/\/protistomics.eu\/martin\/?page_id=136"},"modified":"2019-10-02T14:47:13","modified_gmt":"2019-10-02T14:47:13","slug":"research","status":"publish","type":"page","link":"https:\/\/protistomics.org\/kolisko\/research\/","title":{"rendered":"Research"},"content":{"rendered":"\n<p><strong>Comparative\nGenomics of Diplomonads<\/strong><\/p>\n\n\n\n<p>Some of the microbial eukaryotes are important parasites of humans\nand animals; however, there are many species that are known to be host-associated,\nyet their pathogenicity remains unknown. Comparative genomics is a powerful\ntool for understanding parasitism on a genomic level. Diplomonads are a group\nof microbial eukaryotes that includes medically and economically important\nparasites. Additionally, there are several putative secondarily free-living\nspecies and host-associated commensalic species. We are performing a\ntaxonomically wide, comparative study of diplomonads, including well-studied\nparasites, primarily and secondarily free-living species, and commensals. The\nresults will address some of the fundamental questions of biology, including\nthe mechanisms of reversal from free-living to parasitic lifestyle and whether\nit is possible to predict pathogenicity or host-associated lifestyle based on\ngenomic information.<\/p>\n\n\n\n<p><strong>Diversity\nof Microbial Eukaryotes in the Gut Microbiome<\/strong><\/p>\n\n\n\n<p>We study the\ndiversity of microbial eukaryotes in human gut microbiomes using amplicon\nsequencing by combining several approaches to minimize the presence of host DNA\nin the results. We will take advantage of a current collaboration with the\nInstitute of Experimental Medicine (IKEM, Prague), giving us access to DNA\nmaterial from human microbiomes of healthy individuals as well as individuals\nwith a variety of infections and medical conditions. We will also perform DNA\nsampling from diverse animal hosts. <\/p>\n\n\n\n<p><strong>Microbial\nEukaryotes and Their Interactions with the Gut Microbiome<\/strong><\/p>\n\n\n\n<p>We are using an\nexperimental infection model of a host (mouse or rat) and a microbial eukaryote\n(<em>Blastocystis<\/em>) to identify any\ndetectable changes in transcription levels in the metatranscriptome of the\nmicrobiome (predominantly the prokaryotic part) as well as changes in\ntranscription in the parasite\/commensal and in the host.
